    Enhancement of Bleomycin Sensitivity in Human Lung Cancer Cell Line using Centella asiatica Leaf Extract

    Purpose: To demonstrate the effectiveness of Centella asiatica aqueous extract in augmenting the cytotoxic effect of bleomycin in the adenocarcinoma human alveolar basal epithelial A549 cell line.Methods: The inhibitory effect of bleomycin on A549 cells was determined by incubating the cells for 24 h in different concentrations of bleomycin. The effect of C. asiatica extract on A549 cells was determined after exposing cells to 5, 10, 15, 20, 25, and 30 % (v/v) bleomycin diluted from an initial 10% stock solution. Based on the 50 % inhibitory (IC50) values of bleomycin, 80, 100, and 120 μg/mL concentrations were tested with 15, 20, and 25 % (v/v) of C. asiatica extract.Results: IC50 value of bleomycin on A549 cells was 100 μg/mL, and was reduced to 80 μg/mL when cells were co-incubated with 20 % v/v of C. asiatica extract. The results show that C. asiatica extract reduces the concentration of bleomycin necessary for inhibition of A459 cell growth (IC50). While the IC50 value of bleomycin was 100 μg/ml, it fell to 80 μg/ml when used along with 20 % v/v CA extract (p < 5)Conclusions: This study demonstrates that compounds extracted from C. asiatica may have potential for use as adjuvants in bleomycin cancer chemotherapy.Keywords: Centella asiatica, A549 cells, Bleomycin, Lung cancer, Increased susceptibility, Cell viabilit

    Demonstration of Geometric Landau-Zener Interferometry in a Superconducting Qubit

    Geometric quantum manipulation and Landau-Zener interferometry have been separately explored in many quantum systems. In this Letter, we combine these two approaches to study the dynamics of a superconducting phase qubit. We experimentally demonstrate Landau-Zener interferometry based on the pure geometric phases in this solid-state qubit. We observe the interference caused by a pure geometric phase accumulated in the evolution between two consecutive Landau-Zener transitions, while the dynamical phase is canceled out by a spin-echo pulse. The full controllability of the qubit state as a function of the intrinsically robust geometric phase provides a promising approach for quantum state manipulation.Comment: 5 pages + 3 pages supplemental Materia

    Development of ListeriaBase and comparative analysis of Listeria monocytogenes

    Background: Listeria consists of both pathogenic and non-pathogenic species. Reports of similarities between the genomic content between some pathogenic and non-pathogenic species necessitates the investigation of these species at the genomic level to understand the evolution of virulence-associated genes. With Listeria genome data growing exponentially, comparative genomic analysis may give better insights into evolution, genetics and phylogeny of Listeria spp., leading to better management of the diseases caused by them. Description: With this motivation, we have developed ListeriaBase, a web Listeria genomic resource and analysis platform to facilitate comparative analysis of Listeria spp. ListeriaBase currently houses 850,402 protein-coding genes, 18,113 RNAs and 15,576 tRNAs from 285 genome sequences of different Listeria strains. An AJAX-based real time search system implemented in ListeriaBase facilitates searching of this huge genomic data. Our in-house designed comparative analysis tools such as Pairwise Genome Comparison (PGC) tool allowing comparison between two genomes, Pathogenomics Profiling Tool (PathoProT) for comparing the virulence genes, and ListeriaTree for phylogenic classification, were customized and incorporated in ListeriaBase facilitating comparative genomic analysis of Listeria spp. Interestingly, we identified a unique genomic feature in the L. monocytogenes genomes in our analysis. The Auto protein sequences of the serotype 4 and the non-serotype 4 strains of L. monocytogenes possessed unique sequence signatures that can differentiate the two groups. We propose that the aut gene may be a potential gene marker for differentiating the serotype 4 strains from other serotypes of L. monocytogenes. Conclusions: ListeriaBase is a useful resource and analysis platform that can facilitate comparative analysis of Listeria for the scientific communities. We have successfully demonstrated some key utilities of ListeriaBase. The knowledge that we obtained in the analyses of L. monocytogenes may be important for functional works of this human pathogen in future.     Experimental Evaluation of Fragments from TBM Disc Cutting under Different Load Cases

    The Tunnel Boring Machine (TBM) tunneling process always contains a certain degree of vibrations due to the step broken phenomenon of the cutting tools. Undoubtedly, there is a quite difference in the fragment characteristics which are related to the construction efficiency of TBM under the static load and the combination of static and impact load. In this study, a series of rock breaking tests with a 216 mm diameter disc cutter and marble samples were conducted under different load cases. Based on the Rosin–Rammler distribution curve, the fragments from the cutting tests were also sieved to calculate the absolute size constant (x’) and coarseness index (CI). The relationship between coarseness index, absolute size parameter and the cutting parameters, specific energy, production rate was evaluated. The results show that there is an increasing trend of x’ and CI with the increase of cut spacing and penetration as well as adding impact load component. An overall downtrend in specific energy and upward trend in production rate which are associated with the high efficiency can be observed with the increasing CI and x’. It is believed that the conclusions are of great significance for improving TBM construction efficiency and cutterhead design
